Cloning,Prokaryotic Expression and Antiviral Activity of Chicken Interferon-a Gene

Abstract: The full length of chicken interferon alpha(ChIFN-a) gene was amplified by polymerase chain reaction(PCR)from the genome DNA of Luoman chicken liver,and then sequenced. The amplified gene fragment was 582 by,ofwhich the 489coding region was subcloned into prokaryotic expression vector pQF30. The recombinant plasmidpQEChIFN a was identified by PCR,enzyme digestion and DNA sequencing. SDS PAGE and Western blot showed that a19. 80 kDa fusion protein was expressed in the form of inclusion body with good immunity. After being denatured and re-natured,the activity of inclusion body was detected by inhibiting the cytopathic effect. The activity of recombinant ChIFN“against vesicular stomatitis virus at Vero cell is 1.16 X103 U/ mg.
