The Effect of 2,4-D and KT on Callus Initiation and Somatic Embryogenesis in Cotton Culture

Abstract: An experiment was conduclcd lo study the effect of 2,4-D and KT on callus initiation and somatic cmbryogenesis in collon (Gossypium hirsutum L.) eullurc.The results showed that 2,4-D was essential for callus initiation, and the effect of 2,4-D was enhanced with the supplcmcnl of KT.Both 2,4-D and KT were necessary for the formation of cmbryogcnic cells and cmbryogcnic cell masses.Somatic cmbryogenesis from cmbryogcnic cell masses was promoted with KT, but the action of KT was inhibited with the supplement of 2,4-D.A lot of calli were obtained on MS medium with 2,4-D 0.05mg/L and KT 0.1mg/L after 60 days of culture.The calli were transfercd lo the suspension culture, then many somatic cmbryoids appeared.
