Abstract: TALEN(Transcription activator-like effector nuclease) CRISPR/Cas system(Clustered regularly interspaced short palindromic repeats,CRISPR.CRISPR-associated,Cas) have recently emerged as new technique for targeted genome modifications. They have been successfully applied in many fields such as animals, plants and microorganism. They can generate a double-strand break(DSB).Repair of these DSBs by intracellular DNA repair mechanisms enables to achieve the target gene modification.Compared with traditional method,TALEN and CRISPR/Cas system were stronger specificity, manipulated more easily and higher targeting efficiency. Here we report the construction, function and mechanism of the TALEN and CRISPR/Cas system.

Key words: Gene function, TALEN, CRISPR/Cas system
