Breeding Methods of the New Strain of Genetically Modified Hetao Melon and its Ecological Suitability

Abstract: 1-Aminocyclopropane-1-carboxylic acid synthase(ACS) is the key enzyme in the biosynthasis of ethylene.Total RNA was isolated from ripe Hetao melon fruits.Using oligo(dT)15 as primer,the fruits strand of ACC cDNA was synthesized and a 627 bp DNA fragment was obtained with PCR technique.This ACC cDNA fragment was cloned into site of vector pGA643.Using the pollen-tube pathway to introduce exogenetic genes,then total DNA was isolated from the genetically modified Hetao melon by CTAB method,and was detected for PCR amplification,the result indicated that some seeds had integrated exogenetic genes.Presently,the new strain which had been genetically modified was being done environmental release and generative test.
