小麦转基因再生植株培养体系的优化

Optimisition of the Cultural System of Wheat Transgenic Regenerated Plants

摘要: 以5个基因型不同的小麦幼胚、幼穗作为供试材料,探讨了品种基因型、ABA处理及生根条件等对小麦离体培养的影响。结果表明,小麦品种豫麦18.64为遗传转化的良好受体材料;0.2mg/LNAA和0.8mg/LIAA可促进幼苗生根;在幼胚愈伤组织诱导前14d,用0.5mg/LABA处理可有效抑制出芽率,并有利于促进胚愈伤组织形成和再生能力的保持。

Abstract: The acceptors were obtained f rom 5 wheat cultivars including Yumai 18 64, Yumai 34, Wenmai No. 6,Yumai 70 and 895004.The fac tors affecting the plants regeneration, such as genotype, ABA t reatment and root ing condi tions were studied.The resul t showed tha t Yumai 18 64 was the best acceptor genotype among 5 cul tivars. The hormone propor tions of NAA 0. 2 mg/ L and IAA 0. 8 mg/L could ef f iciently improve root dif ferentiation. Meanwhi le i t was found in the experiments that immature embryo germination was not only inhibi ted, but embryonic cal lus development could also be significantly improved by culturing callus on the medium c ontaining ABA(0. 5 mg/ L) for two weeks.
