分子标记辅助选择改良水稻恢复系R389及其杂交种稻瘟病抗性

Improving Blast Resistance of Rice Restorer 389 and Its Hybrid Through Molecular Marker-assisted Selection

摘要: 为了定向改良籼稻恢复系R389及其配制的杂交种的稻瘟病抗性,利用广谱持久抗稻瘟病基因Pi9设计特异性功能分子标记Ins1-1,用于开展分子标记辅助选择连续回交育种。室内接种抗菌圃分析结果表明,Pi9基因供体亲本75-1-127对20份供试稻瘟病菌中的18个小种表现出高水平抗性,而受体亲本R389对其中11个小种表现出抗性,双亲间抗性表现与抗菌谱差异明显,抗性频率分别为90.0%,55.0%;供体亲本75-1-127在自然稻瘟病病圃的苗瘟和穗瘟抗性表现均为高抗,R389的苗瘟和穗瘟抗性表现均为高感。Ins1-1是一个基于PCR的显性DNA标记,在两亲本间表现出明显且稳定的多态性;在Pi9供体亲本75-1-127和改良获得的R389群体及其配制的杂交种基因组中均扩增出一条大小为513 bp的稳定条带,而在受体亲本R389及两优389基因组中均无扩增产物;Ins1-1标记基因型对稻瘟病抗性表型的辅助选择效率达100%。从26个含Pi9纯合等位基因的BC6F3株系中筛选出一个高抗苗瘟和穗瘟、田间性状与受体亲本R389接近的改良恢复系R389-Pi9,并用它与温敏不育系P88s配制出高抗稻瘟病的两系杂交种两优389-Pi9;田间小区栽培试验结果表明,改良恢复系及其所配置的杂交种的全生育期、株高、结实率、千粒质量等主要农艺性状与相应对照相比均无显著差异,表现为遗传背景基本一致,实现了定向改良恢复系R389及其杂交种稻瘟病抗性的目标。

Abstract: To directionally improve blast resistance of the indica rice restorer R389 and its hybrid,a functional DNA marker Ins1-1 was developed according to the broad-spectrum and durable blast resistance gene Pi9, and molecular marker-assisted selection(MAS) consecutive backcross breeding were implemented. Resistance responses and resistance spectra to twenty tested M.oryzae isolates were quite different between the Pi9 gene donor 75-1-127 conferring resistance to eighteen isolates and the receptor R389 which showed resistance to only eleven isolates,and the resistance frequencies were 90.0% and 55.0%,respectively in the greenhouse inoculation assay. Accordingly,75-1-127 showed high-level blast resistance during seedling stage and panicle developing stage but R389 was highly susceptible in field nursery. Ins1-1 was a PCR-based dominant DNA marker,showed obvious and stable polymorphism between two parents,by amplifying a 513 bp band from the genome of donor 75-1-127,the pure line of R389-Pi9 and its hybrid,but no product from that of the receptor R389 and its hybrid. Selection efficiency by Ins1-1 genotype for blast resistance phenotype reached 100%. An improved restorer line R389-Pi9 was bred from the twenty-six BC6F3 family lines,which showed similar field characteristics but much better blast resistance comparing to R389,and its two-line hybrid Liangyou 389-Pi9(P88s/R389-Pi9) was created which showed high level seedling and panicle blast resistance in field nursery. Field cultivation assay showed that the improved pure line R389-Pi9 and its hybrid Liangyou 389-Pi9 showed no significant difference to corresponding contrasts in main agronomic traits such as growth period,plant height,seed setting rate and 1000-grain weight,implying similar genetic background and improved blast resistance were achieved.
