1Dx5在新冬26小麦中的遗传转化及分析

1Dx5 Genetic Transformation and Analysis in 26 New Winter Wheat

摘要: 为了对转1Dx5基因小麦的T1进行遗传分析,将优质高分子量麦谷蛋白亚基(HMW-GS)基因1Dx5转入新疆耐盐小麦品种新冬26,在蛋白质水平上经SDS-PAGE检测,有37粒小麦种子1Dx5基因表达。经过继代培育,得到37个T1转基因株系,SDS-PAGE分析T1转基因小麦籽粒的HMW-GS组成,结果表明:在蛋白质水平,HMW-GS的组成在T1小麦种子中有3种表型:表型Ⅰ,外源1Dx5基因没有表达;表型Ⅱ,外源1Dx5基因表达;表型Ⅲ,内源1Dx2基因没有表达。这3种表型出现的概率分别约为34.3%,45.9%和19.8%。该研究成功获得了新的HMW-GS组成的小麦,为选育优质小麦提供分子依据。

Abstract: In order to carry out genetic analysis of T1 generation of transgenic wheat with 1Dx5 gene,the Xinjiang winter common wheat Xindong No.26 were transformed with high-molecular-weight glutenin subunits (HMW-GS) gene 1Dx5,detected by SDS-PAGE,was found expression of 1Dx5 gene in 37 wheat seeds at the protein level.After subculture cultivation,37 T1 generation transgenic lines were obtained.SDS-PAGE analysis of T1 generation transgenic wheat seeds,the results showed that the composition of HMW-GS were found three phenotypes at protein level,phenotypic Ⅰ,the foreign 1Dx5 gene did not express;phenotypic Ⅱ,the foreign 1Dx5 gene express;phenotypic Ⅲ,endogenous 1Dx2 gene did not express.The probability of these three phenotypes were approximately 34.3%,45.9% and 19.8%,respectively.The study successfully obtained new types of HMW-GS composition,providing molecular evidence for the selection of high quality wheat.
