基于紫娟茶叶片转录组的SSR引物开发

SSR Primers Development Based on Zijuancha Leaves' Transcription Group

摘要: 以稀有茶树品种紫娟茶为研究对象,进行SSR引物开发的研究,从紫娟茶叶片转录组中筛选重复基元最多的SSR,设计并筛选紫娟茶的SSR引物。通过从已测序的紫娟茶叶片转录组中设计SSR引物共87对,通过对紫娟茶以及随机采摘的3份不同品种茶树提取DNA后进行PCR扩增,1%琼脂糖凝胶电泳分析,在紫外凝胶成像仪中观察拍照,判断是否扩增出目标条带。通过试验筛选出可以扩增出清晰目标条带的引物63对,有效率达72.4%,其中有33对表现出多态性,占有效SSR引物的52.4%。结果表明,从紫娟茶叶片转录组中设计引物是可行的。下一步研究将从这63对引物中开发紫娟茶的特异引物。

Abstract: To develop SSR Markers from a rare tea variety Zijuancha,Zijuancha leaves' transcriptome which had the highest number of SSR repeats were used to design and select Zijuancha SSR Markers.In this study,designed primers from sequence of transcription group of Zijuancha's leaves,and got 87 primer pairs.Took DNA from Zijuancha and 3 other tea accessions,PCR by each primer,and analyzed in 1% agarose gel electrophoresis.Results showed 63 primer pairs could amplify bands,with the effective amplification rate of 72.4%.33 primer pairs demonstrated the polymorphic characteristics,accounting for 52.4% of all effective primer pairs.According to this study,to designed primer pairs from sequence of Zijuancha transcription group was viable.To picked up specific primers of Zijuancha from those 63 primer pairs will be performed in the next study.
