八眉猪DQA基因编码区多态性及生物信息学分析

Cloning and Bioinformatics Analysis on Coding Regions of DQA Gene in Bamei Pigs

摘要: 采用克隆测序的方法对八眉猪 DQA 基因编码区进行多态性分析,并利用生物信息学方法预测八眉猪 DQA 基因的理化特性及其编码产物的结构功能。结果显示:八眉猪 DQA 基因编码255 个氨基酸,编码区序列有14 个碱基变异,11 个氨基酸变异,外显子2区域表现出丰富的多态性。DQA 基因编码产物是一种不稳定的水溶性蛋白,二级结构为混合型,三级结构由α-螺旋、β-折叠、β-转角和无规则卷曲组成。 蛋白质功能预测结果显示,信号转导、受体、胁迫应答、免疫应答和生长因子的几率较高。研究结果为八眉猪的抗病分子育种和经济性状相关基因筛选提供理论依据。

Abstract: Polymorphism of DQA gene coding regions in Bamei pigs were analyzed by cloning and sequencing, physicochemical characteristics, protein structures and functions were predicted through the bioinformatics tools.The results showed that DQA gene encoded 255 amino acids, there were 14 nucleotide and 11 amino acid substitutions in DQA gene coding regions, and it was showed high polymorphism in exon2 region.Encoding product of DQA gene was a kind of instable and soluble protein, the putative secondary structure of DQA was mixed, and putative protein tertiary structure was consisted of alpha helix, extended strand, beta turn and random coil.The putative result of DQA protein function showed that the odds of signal transducer, receptor, stress response, immune response and growth factor were higher.These results will provide a basis for the study of molecular breeding in disease resistance and economic traits related gene screening.
