中国荷斯坦牛BOLA-DRB3外显子2的PCR-SSCP多态性分析

摘要: 通过PCR-SSCP法对中国荷斯坦牛BoLA-DRB3基因的外显子2进行多态性分析.在223个个体中共获得10种等位基因,其中D等位基因频率最高为0.228 7,A等位基因频率最低为0.013 ;等位基因型18种,其中FG基因型频率最高,为0.12 6,其次为HH,基因型频率为0.112 1;ML基因型频率最低,为0.004 .另外分析发现,在不同家系中,DRB3基因外显子2等位基因具有特异性,等位基因的类型和频率存在比较明显的差异.差异显著性分析表明,D等位基因对30 d产奶量和30 d乳蛋白量有明显的负效应,ME基因型个体30 d乳脂量显著低于其他个体.结果表明中国荷斯坦牛的BoLA-DRB3外显子2具有丰富的多态性,并具有家系分布的特异性.

Abstract: In the present work,genetic polymorphisms of BoLA-DRB3 * exon2 gene in 223 Chinese Holstein were detected by PCR-SSCP. In total,18 SSCP patterns were identified and the frequency of each pattern within the herd was determined. The frequency of pattern FG was the highest which was 0. 125 6,the next frequency of pattern HH was 0. 112 1,the frequency of pattern ML was the lowest which was 0. 004 5. We found that SSCP patterns of different families were specific. Differences of significance analysis indicates that,D alleles has a significant negative effects on 305 days milk yield and 305 days milk protein content,305 days milk fat content of ME genotype significantly lower than other individuals. Preliminary analysis result revealed high genetic polymorphism of exon2 region of BoLA-DRB3 gene in Chinese Holstein.
