小麦新种质N95175抗白粉病基因的SSR分析

摘要: 为明确小簇麦新种质N95175抗白粉病基因的遗传效应和基因位点,采用常规分析法结合SSR技术进行抗性基因的遗传分析和分子标记研究。抗性基因常规分析结果表明,N95175分别与高感白粉病普通小麦品种陕160和陕优225两个杂交组合的F1均现高抗,F2抗感植株比例分别为115∶43和111∶48,经χ2检验,符合3∶1的显性单基因孟德尔遗传分离比例,即该抗白粉病基因为显性单基因遗传。利用208对小麦微卫星引物对N95175×陕优225的F2抗感分离群体分析结果表明,Xgwm570和Xwmc553均与抗白粉病基因连锁,遗传距离分别为13.38和12.03 cM。Xg-wm570和Xwmc553两者之间在两群体中的遗传距离为3.74 cM。利用两引物对N95175×陕160组合F2代进行标记验证分析,分析结果与接种后调查结果符合率为89.24%。根据Xgwm570和Xwmc553在小麦染色体的位置,将N95175的抗白粉病基因定位在6A染色体上。

Abstract: To definitude genetic effects and gene loci of powdery mildew resistance gene in new wheat2haynalidiavillosa germplasm N95175,conventional analyze and SSR technology was used to analyze heredity and molecular markersof resistant gene. The results showed that F1of N95175 × Shaan160 and N95175 × Shaanyou225 were highly resistant topowdery mildew. The ratios of resistance plants to susceptible plants in F2generation were 115∶ 43 and 111∶ 48,respec2tively. The segregation ratio fitted expected 3∶ 1,so it suggested that powdery mildew resistance of N95175 was controlledby a dominant gene.By SSR analysis with 208 pairs of primers,two markers Xgwm570 and Xwmc553 generated polymor2phic DNA fragments between the resistant and susceptible plants DNA pools. By analyzing the polymorphic markers inthese segregating populations,Xgwm570 and Xwmc553 were linked to the resistance gene with genetic distance of 13138and 12103 cM. The genetic distance between marker Xgwm570 and Xwmc553 was calculated as 3174 cM.By using twomarkers,the marker certification was analyzed in N95175 × Shaan160 F2generation,the according rate was 89124 % com2pared with inoculation. According to chromosome loci of Xgwm570 and Xwmc553,powdery mildew resistance gene inN95175 was located on wheat 6A chromosome.
