水稻GluB_1启动子克隆及序列分析

Cloning and Sequencing of the Glu B-1 Promoter from Rice

摘要: GluB1是水稻种子谷蛋白的编码基因,是种子成熟过程中,由GluB1启动子调控,特异地在胚乳中表达的蛋白。以水稻基因组DNA为模板,通过PCR扩增得到Glu B1启动子片段,将其克隆在pGEMT Easy载体上进行序列分析。结果表明:获得的启动子片段的大小为2 293 bp,与已报道的该启动子序列相比较,其核苷酸序列同源性为99.2%。该启动子区域含有ACGT基序、AACA基序和GCN4基序等胚乳特异表达必需元件。

Abstract: The GluB_1 promoter was amplified from Oryza sativa genome by polymerase chain reaction and cloned into pGEM-T Easy vector.Sequence analysis showed that the obtained promoter fragment 2293 nucleotides and showed homology of 99.2% with the reported Glu B-promoter.Also,the obtained promoter fragment includes the three cis-regulatory elements,ACGT-motif,AACA-motif and GCN4-motif,which are necessary to mediate endosperm expression of the GluB_1 glutelin gene.
