小枣发育枝愈伤组织类型及细胞学观察

摘要: 在金丝小枣发育枝离体培养过程中,不同种类和浓度的激素及光照条件对诱导愈伤组织的质量有明显影响。2,4-D和暗培养易诱导出疏松的Ⅱ型愈伤组织,IBA及光照培养则有利于致密的Ⅰ型愈伤组织形成。Ⅱ型愈伤组织未分化出芽,而Ⅰ型具有较强的再分化能力。继代繁殖时,2,4-D浓度超过1mg/L将导致Ⅰ型愈伤组织大量转化为Ⅱ型。不同类型愈伤组织的染色体倍性变异明显不同,再生植株无性系根尖细胞染色体倍性则相对稳定。

Abstract: It was found in the in vitro culture of the callus from the growing branch of the “Jinsi Xiaozao” jujule ,that stimulants at different concentration and lighting significantly affected the quality of the callus.The use of 2,4 D in dark culture helped the induction of loose type Ⅱcalli,while IBA and lighting helped the formation of compact type I calli .The type I calli showed no capacity of differentiation ,while the type Ⅱcalli was found stronger in differentiation .During propagation 2,4 D at a concentration higher than 1mg/L resulted in the massive conversion of type I into type Ⅱcalli .Calli were found different in the variability of chromosome ploidy .The cells from the root tip of the regenerated plantlets were found relatively stable in their chromosome ploidy .
