三个棉种茎尖培养再生植株培养基的研究

摘要: 海岛棉、陆地棉、中棉的茎尖(5-10mm)培养在改良MS附加IAA和Kt的培养基上,均诱导产生了完整小植株,由此为繁殖植茎尖无性系提供了可能性。改良MS培养基不但适用于棉花二倍体茎尖培养,而且适用于棉花单倍体茎尖培养,这为单倍体的快繁提供了可能性。棉株发育初期的茎尖和腋芽较现蕾后的茎尖和腋芽更易于生根,根、茎、叶分化也好。日温32℃、夜温28℃比试验的其它温度更有利于棉花生根和茎、叶分化。

Abstract: Shoot apex(5-10mm in lenght)of Sea Island Cotton(Gossypium Barbadense L.),Upland Cotton(Gossypium Hirsutum L.)and Tree Cotton(Gossypium Arboreum L.)were cultured on improved MS medium supplemented with IAA and KT,all resulting in formation of complete plantlet.This way provided the possibility of micropropagation of cotton clones derived from the shoot apex.The improved MS culture medium is applicable not only to the cultivation of cotton diploid with shoot apex,but also to the cultivation of cotton haploid with shoot apex.It provided possidility for the fast breeding of haploid.Shoot apex and axillary bud in the early stage of the development of cotton plandet are easier to strike roots than those in the bud and boll sqaring stage,and their roots,shoot and leaf differentiation are also better.The temperature 32℃ in daytime and the temperature 28℃ in nighttime aremore favorable to root striking and differentiation of shoot and leaf than other temperatures in the experiment.
