Effect of Culture Time and Inoculum Density on Reproduction and Morphometrics of Pratylenchus coffeae in Carrot Disk Cultures

Abstract: Effect of culture time and inoculum density on reproduction and morphometrics of Pratylenchus coffeae was evaluated with carrot disk culture method. Each carrot disk was inoculated 25 P. coffeae females, the nematode population increased with increasing culture time. The reproductive rate of nematodes is 6.24 in the first three-weeks and 8.99 in the second three-weeks. Afterwards the nematodes reproductive rate increased sharply and reached to 17.88 from the 6th week to 9th week; after the 9th week reproductive rate of nematode decreased to 1.42. The nematode population increased significantly in all the treatments of the density comparing experiment, but its reproductive rates were not significantly influenced by the nematode densities. Ten female and male nematodes were randomly picked from different density treatments, and their morphometrics were measured. All the morphometrics of female and male did not show significant differences among different density treatments except the b value (length of body/distance from anterior end to pharyngo-intestinal valve) of the female.
