Effects of Phospholipase C Inhibitor U-73122 on Conidial Germination and HT-toxin Activity of Setosphaeria turcica

Abstract: Phospholipase C(PLC)could catalyze the hydrolysis of phospholipid molecules on cell membrane and generate two second messenger molecules IP3 and DAG, which activated Ca“signal pathway and protein kinase C re— spectively.It had been proved that PLC modulated pathogenicity and development of plant pathogenic fungi.In this research, U-73 1 22, the specific inhibitor of PLCs, was used to study functions of PLCs in Setosphaeria turcica prelimina— ry.U一73122 showed significant inhibition effects on hyphal growth and conidial germination.The higher was the inhibitor concentration, the lower was the germination ratio of spores.U-73 122 caused the germ tube shortening and cell swelling irregularly.In addition, U一73 1 22 could also obviously decrease the biological activity of HT-toxin.It was indi— cated that PLC might regulate hyphal growth and conidial germination by controlling the cell polarity, and also involved in the biosynthesis of HT—toxin in Setosphaeria turcica.
