Cloning and Bioinformatic Analysis of Metallothionein Gene(MxMT2)in Malus xiaojinensis

Abstract: Ametallothione in genewas clonedby RT-PCR andsplicing of 27 cDNA sequences thatwereidentif ied by differentia lscreening of the subtracted cDNA libraryfrom Malusxiao jinensis Chenget Jiang, which was named MxMT2 (AccessionNo.GQ906589).Thefull-lengthcDNAofMxMT2 is610 bp, including the openreading frame (ORF) of 240 bp.It encodes a protein of 80 aminoacids, with amolecularweight of 7.74 kDa.The MxMT2 contained the conserved domains sharedin all known MT, which has 14 Cysresidues(cys)that have conserved sequence position at Nterminal and Cterminal of protein, rankingin the forms of Cys-Cys, Cys-X-Cys and Cys-X-X-Cys.Thephy logenetictree suggested that MxMT2 wascloser to the MTofsandpear, almondandapricot, but fartherwith the MT of europeanbeech, hankow willow and adzuki bean.
