Molecular Cloning of Aquaporin1 cDNA and Multi-tissue mRNA Expression in Monopertus albus

Abstract: Monopertus albus Aquaporin 1 ( AQP1 ) cDNA was cloned to investing its structure, evolution, and expression in different tissues by RT-PCR and RACE method.The result shown that the AQP1 cDNA is composed of a 163 bp 5'UTR, 182 bp 3 'UTR and a 1 134 bp nuclotides with a 952 bp nucloetide open reading frame encoding a protein of 263 amino acids.Protein sequence alignment showed that the highest similarity with the European eel ( Monopertus albus) AQP1 (87% ) and the other is 42.9% , 61 .2% , 46.1% , 42.9% , 53 .9% , 47.9% , 41 .5% ,40.1 % , 33 .2% , 30.8% , 33 .2% .Bioinformatics analysis revealed that the protein whose calculated Molecular weight is 27.8 kDa and theoretical isoelectric point is 7.945 was hydrophilic in most region and had rich B cell antigencity positions.The survey about multiple tissue expression of AQP1 gene in eel revealed that the AQP1 mRNA is mainly expressed in ovary and the concentration of mRNA in other tissues is very small.
