Representational Difference Analysis of DNA Sequences Related to Cartap Resistance in Diamondback Moth,Plutella xylostella L.

Abstract: The genomic difference between Plutella xylostella L.susceptible strain and cartap resistance near isogenic line strain was preliminarily analyzed by representationaLdifference analysis ( RDA)technique.In RDA system, the driver amplicon is susceptible strain while the tester amplicon is cartap resistance near isogenic line strain.After three rounds of subtractive hybridization, the different products were obtained and their sizes were about 150-300 bp.Comparisons between our sub-clone sequencing results and GenBank show that only one sub-clone sequence has P450 gene homology, other sub-clone sequences show no homology to knew gene.Preliminary judgment revealed that these segments might be new gene sequences.
