Abstract:
Inactivated oil emulsion vaccine against ND was studied and prepared successfully,by using NDV Lasota strain as seed virus,formalin as inactivating agent,No 7 white mineral oil as adjuvant, Tween-80 and span-80 as emulsifiers, merthiolate as preservative. This oil-emulsion vaccine, of a double emulsion type (W/O/W), had particles about 1μm in diameter. The vaccine was thin emulsion with the viscosity≤1 second,it was easy to inject. Under 4~8℃,the vaccine can be preserved for over one year. Potency tests indicated the dosage was ≥70PD50 The earliest immunity was produced at 12 days after inoculation (100% prortection). Practically, the preferred dosage was 0. 5ml per bird for birds within 2 months of age, and 1. 0ml per bird for the birds older than 2 months of age. The vaccine was safe for chickens of any ages, and had no adverse effects. When combined with live ND vaccines, it could induce high HI antibody for chicks of any ages. Immune periods lasted 2. 5 to 3 months for chickens and more than 6 months for adult birds. The viscerotropic velogenic ND and atypical ND can be prevented effectively by using this vaccine.
