表皮生长因子对牛输卵管上皮细胞生长、增殖和凋亡的影响

Influence of Epidermal Growth Factor on Cell Proliferation Cycle and Apoptosis of Cultured Bovine Oviductal Epithelial Cells in vitro

摘要: 研究不同浓度的表皮生长因子对牛输卵管上皮细胞生长、增殖和凋亡的影响,目的是建立高质量的牛体外胚胎共培养细胞系。牛输卵管上皮细胞体外培养技术结合血球计数法检测细胞增殖,并利用流式细胞仪进行细胞周期和凋亡的检测,通过Lysis软件对数据进行分析。结果表明,EGF在0~50 ng/mL浓度范围内,其促增殖、抑制凋亡作用随浓度的增加而增大,呈剂量依赖性;当浓度持续增高时,其促增殖、抑制凋亡能力降低;此外,S期细胞数量明显增加,G2-M期细胞数量有下降趋势。表皮生长因子在一定浓度范围内具有促进牛输卵管上皮细胞增殖和抑制其凋亡的作用,并使其细胞周期发生变化。

Abstract: To investigate the effect of epidermal growth factor ( EGF) on proliferation, cell cycle and apoptosisof cultured bovine oviductal epithelial cells and provide foundation for further study of system of co-culturing bovineembryo in vitro. The cultured bovine oviductal epithelial cells were treated with different dose of EGF. The effects ofbovine oviductal epithelial cells proliferation, cell cycle and apoptosis were measured by blood counting assay andanalyzed by FCM ( flow cytometry) and Lysis software. In the definite range of EGF (0 - 50 ng /mL) , the effect oncells increased with raise of the EGF concentration showed dose-dependently. With the concentration improved, theabilities of promoting proliferation and inhibiting apoptosis had decreased. Besides, cell quantities increased obviously at S phase while declined slightly at G2-M phase. In the definite range of concentration, the epidermal growth factor could promote the proliferation and inhibiting apoptosis of the cultured bovine oviductal epithelial cells, moreover, the cell cycles had been altered after adding EGF.
