黄芩苷抗热应激小鼠输卵管氧化损伤的研究

Effect of Baicalin on Oxidative Damage of Mouse Oviduct Induced by Heat Stress

摘要: 为降低热应激引起的动物机体活性氧(ROS)增加,减少组织氧化损伤和细胞过度凋亡。利用小鼠模型研究热应激对输卵管组织和细胞的损伤作用以及黄芩苷(Bai)对损伤的保护作用。将小鼠分为对照(Ⅰ)组;Bai(Ⅱ)组;热应激(Ⅲ)组和热应激加Bai(Ⅳ)组。用试剂盒检测组织丙二醛(MDA)含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活力;用Western Blot检测Kelch样环氧氯丙烷相关蛋白-1(Keap1)、核因子E2相关因子2(Nrf2)、苯醌氧化还原酶(NQO1)和血红素氧合酶-l(HO-1)蛋白表达;用免疫组化方法检测凋亡因子Apaf-1和Caspase-9蛋白表达。结果显示,与对照组相比热应激后小鼠输卵管组织MDA含量显著增加(P<0.05),CAT和GSH-Px活力显著降低;同时,Nrf2、Keap1蛋白表达量显著升高(P<0.05),凋亡因子Apaf-1和Caspase-9蛋白表达明显增加。注射黄芩苷后可显著(P<0.05)降低小鼠输卵管组织MDA含量和Nrf2和Keap1蛋白表达量,降低凋亡因子Apaf-1和Caspase-9蛋白表达,并且显著增加(P<0.05)SOD、CAT和GSH-Px活力。结果表明,黄芩苷能降低热应激小鼠输卵管组织的氧化损伤和细胞凋亡,其机制可能与激活抗氧化信号通路Keap1-Nrf2/ARE有关。

Abstract: In order to reduce the heat stress caused by animal body reactive oxygen species (ROS) increased, reducing tissue oxidative damage and cell apoptosis.The effect of heat stress on the oxidative damage of oviduct tissues was studied by using the mouse model, and the protective effect of baicalin on the oxidative damage of the oviduct was investigated. The mice were divided into control(group Ⅰ), Bai(group Ⅱ),41℃ heat stress (groupⅢ) and 41℃ heat stress+Bai (group Ⅳ). Malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) were detected by kits, respectively. The expression of Nrf2, Keap1, HO-1 and NQO1 proteins were detected by using Western Blot and the expressions of Apaf-1 and Caspase-9 were detected by immunohistochemistry method. The results showed that compared with the control group, the content of MDA in oviduct tissue was significantly increased (P<0.05) and the activities of CAT and GSH-Px were significantly decreased after heat stress.The expression of Nrf2 and Keap1 proteins were significantly increased(P<0.05). Also Apaf-1 and Caspase-9 proteins were significantly increased as compared with control group. However, compared with group Ⅲ, baicalin significantly (P<0.05) decreased the content of MDA and the expression of Nrf2 and Keap1 proteins. The apoptotic related proteins of Apaf-1 and Caspase-9 were also decreased. The activities of SOD, CAT and GSH-Px increased significantly (P<0.05).These results indicate that baicalin can reduce the oxidative damage and apoptosis of oviduct tissue of mouse under heat stress, and its mechanism may be related to activation of antioxidant signaling pathway of Keap1-Nrf2/ARE.
