烤烟不同叶位叶片有机酸含量的差异分析

Effects of Transgenic Insect-resistant Cotton on Development and Metabolite Content of Cotton Aphid Aphis gossypii

摘要: 采用硫酸甲酯化前处理,结合气相色谱-质谱(GC/MS)分离分析技术,研究了烤烟不同叶位叶片(L8-L12,即从底部第8位叶向上到第12位叶)有机酸含量的差异。GC/MS定量分析结果表明:烤烟不同叶位叶片中,苹果酸、γ-戊酮酸、柠檬酸、丙二酸、乙二酸、亚麻酸和软脂酸含量相对较高,是烤烟中主要的有机酸成分;并且不同叶位叶片有机酸、苹果酸与其他有机酸含量差异明显;不同叶位叶片有机酸总量表现为:L8>L9>L12>L10>L11;L8-L12叶片中,非挥发性有机酸、高级饱和脂肪酸、高级不饱和脂肪酸总量分别在有机酸总量中所占比例范围是:88.49%~94.38%,3.01%~5.10%,2.61%~6.40%。不同叶位叶片有机酸含量差异分析结果表明:L10和L12位叶有机酸含量适中,具有优质、高档烟原料的特质。

Abstract: In recent years,the population densities of non-target pests in transgenic Bt cotton fields were obviously increased.Cotton aphid Aphis gossypii(Homoptera: Aphidae) was one of the seriously non-target pests in transgenic Bt cotton field.One A.gossypii strain was respectively reared on transgenic Bt cotton line SGK321,expressing Cry1Ac insecticidal protein,and its transgenic parent cotton cultivar Shiyuan321 for more than forty generations in the laboratory.The contents of Bt protein,carbohydrate,lipid,protein and life table parameters in A.gossypii,were measured and compared on two host plants SGK321 and Shiyuan321.The results showed that very low concentration of Bt protein detected in A.gossypii reared on Bt cotton.The transgenic Bt cotton SGK321 had no significant impacts on mortality,reproduction and life-table parameters of A.gossypii.The contents of carbohydrate,lipid and protein in A.gossypii,fed on transgenic Bt cotton for a long period of time,were also not significantly affected compared to fed on control cotton.Datum indicated that transgenic Bt cotton SGK321 had no direct acute toxic effects on A.gossypii in the long term.
