农杆菌介导法将LFY基因导入嘎拉苹果的研究

Study on Transferring LFY Gene into Gala Via Agrobacterium tumefaciens Mediation

摘要: 以嘎拉苹果叶片为外植体,利用GUS瞬时表达率,对农杆菌介导法转化苹果的因素进行优化。结果表明:卡那霉素(Km)选择压在30 mg/L时,外植体预培养2~3 d后以OD600=0.3~0.5农杆菌菌液浸泡3~5 min,共培养3 d,延迟筛选3 d可明显提高M26叶片转化效率,GUS阳性率达50%。PCR检测初步证明,LFY基因已整合到苹果再生植株中。

Abstract: The factors affecting apple transformation mediated by Agrobacterium tumefaciens, such as co-cultivation time, bacterium density et al., had been studied through GUS transient expression. Apple of Gala was transformed by Agrobacterium tumefaciens EHA105 carrying LFY gene from Arabidopsis. It was proved that the target LFY gene had been integrated into the genome of regenerated plants by PCR analysis.
