普通小麦细胞质雄性不育保持系mtDNA基因文库的构建

Construction of mtDNA Library of CMS Maintainer of Common Wheat

摘要: 以普通小麦细胞质雄性不育保持系75-3369B为材料,分别用限制性内切酶EcoRⅠ和MboⅠ对75-3369B的mtDNA进行完全酶切和部分酶切,构建了两个基因文库。其中用EcoRⅠ酶切得到的库(以下简称ME库)由350个重组子构成;MboⅠ酶切所构建的库(以下简称MM库)由450个重组子构成,MM库的库容量是98.12%。同时,讨论了两种建库方法的优缺点。

Abstract: Cytoplasmic male sterility(CMS)line 75-3369A's maintainer 75-3369B of common wheat was studied. mtDNA was digested completely with EcoRⅠ and partly with MboⅠ and two gene libraries(ME and MM)of mitochondrial DNA(mtDNA)of 75-3369B was constructed. ME consisted of 350 clones and MM contained 450 clones. MM library included 98.12 percent of the mitochondrial genome. The advantage and disadvantage of the two methods of gene library construction were also discussed.
