十字花科根肿病接种技术及发病条件研究

摘要: 为了明确接种方法、环境和基质条件等对十字花科根肿病发病的影响.研究比较了不同接种方法、肿根保存形式、光源及光周期、基质类型及基质pH值等对大白菜根肿病发病程度的影响,以建立一套简便、稳定的大白菜根肿病接种技术及品种抗病性鉴定体系.结果表明,大白菜种子在培养皿中催芽7 d后,采用浸根接种法,不仅节省菌源,而且操作简单,发病稳定;自然光照与稀土灯补光结合16 h光周期条件下,大白菜根肿病发生最严重,且发病均匀,为十字花科蔬菜根肿病发生的最佳光照环境;草炭和蛭石基质有利于根肿病的发生,砂土和壤土对根肿病发生有抑制作用;根肿病在pH 值5.0和pH 值7.0时均可发病,但酸性环境有利于根肿病的发生;另外,肿根干燥保存不影响病原菌活力,可作为实验室保存肿根的推荐使用方法.

Abstract: To know the effect of inoculation methods,substrate and matrix conditions on the incidence of clubroot disease.A stable inoculation technique was developed to inoculate Chinese cabbage with Plasmodiophora brassicae,which could be adoptable in evaluation of resistance of cruciferous to clubroot.The inoculation methods,clubroot storage methods,light sources,photoperiods,substrates and pH environment of culture system etc were studied to compare the affection to disease incidence of clubroot.The results showed that root soaking inoculation,with 7-day-old seedlings cultured on Petri dish,caused more severe and stable symptoms,and was much simple and efficient.Natural light supplemented by artificial light with a 16 h photoperiod was recognized as the best light source for inoculation.Compared with soil and sand,peat and vermiculite were the optimal matrixes for clubroot development.Acid matrixes were favorable for severe attack of the disease,whereas neutral matrixes were less favorable.In addition,the clubroot maintained high pathogenic vitality after air-dried at room temperature and preserved at dry environments,which demonstrated that clubroot drying storage could be recommended as a preservation method in laboratory.
