聚谷氨酸对小鼠钙吸收的影响

摘要: 为获取γ-PGA高产突变株,确定聚谷氨酸对钙溶解性和对小鼠体内钙吸收的影响。使用紫外照射纳豆芽孢杆菌诱导突变,根据形态特征变化筛选、培养突变株。分离纯化不同突变株的γ-聚谷氨酸(γ-PGA)并测定产量。筛选出γ-PGA高产的突变株并测定其遗传稳定性。测定γ-PGA在体外和体内对钙溶解性的影响以及γ-PGA对小鼠骨矿物质含量的影响。筛选出了具有稳定遗传性的γ-PGA高产突变株Z-29,其γ-PGA的产量大于18 g/L。结果表明,在体外,γ-PGA能促进钙的溶解性;在体内,γ-PGA也能促进小鼠小肠钙的吸收,并能增加小鼠骨钙和骨磷的含量。筛选出了纳豆芽孢杆菌γ-PGA高产的突变株,证明了γ-PGA在体内外都能增加钙的溶解性,这为更深入地研究γ-PGA的功能和生产应用提供了依据。

Abstract: The purpose of this study is to obtain mutant strain with high yields of γ-PGA and determine polyglutamic acid effect on calcium absorption in mice.Bacillus natto was irradiated by ultraviolet to induce mutation.The mutant strains were selected according to morphological changes.Then,the mutant strains were cultivated.The γ-poly glutamic acid (γ-PGA) of different mutant strains were separated and purified to determine its production.The mutant strain which had high yields of γ-PGA was selected and determined its genetic stability.The effects of the calcium solubility by γ-PGA in vitro and in vivo were measured as well as the bone mineral content in mice.Experiment screened out a mutant strain,Z-29,with stable high yields of γ-PGA greater than 18 g/L.The experimental results showed that γ-PGA promoted calcium solubility in vitro and in mice small intestine.Moreover,it increased bone calcium and bone phosphorus content in mice.This experiment gained Bacillus natto mutant strains with high yields of γ-PGA and proved that γ-PGA can increase the solubility of calcium in vitro and in vivo.It provided the basis for further research and application in production.
