转基因抗虫棉Bt基因与npt II基因的遗传与表达

摘要: 采用花粉管通道法将含有Bt杀虫基因的pG4AB质粒导入受体棉花中2中,通过卡那霉素筛选,Bt免疫检测条检测,PCR扩增对转基因抗虫棉的各个自交世代进行跟踪研究。结果表明,具有卡那霉素抗性的单株能扩增出Bt基因,而且均有Bt杀虫蛋白表达。Bt基因与npt II基因均整合进了棉花基因组中,相互之间紧密连锁遗传,均稳定表达,因此报告基因npt II可用于研究Bt基因的遗传情况。

Abstract: Plasmid p G4AB containingBtgene was transferred into cotton Zhong 23 via pollon tube pathway.The Kanamycin detection and the expression ofBtICP,PCR were used to analyze the self-pollination generations from transgenic cotton Zhong 23.The results were as follows.ExogenousBtgene was amplified and expressed in the Kanamycin-resistance transgenic cotton.TheBtgene andnpt IIgene were inserted into the transgenic cotton genome,they were linked and inherited stably in different generations.npt IIgene can be used in inheritance analysis ofBtgene.
