马铃薯脱毒微型种薯打破休眠的研究

A Study on Dormancy Breakage in Virus-free Potato Minituber

摘要: 采用激素酶联免疫吸附法(ELISA)检测贮存在3种温度下,4个不同处理的微型薯内源激素GA3、IAA和ABA含量的变化及对微型薯休眠的影响;微型薯经外源激素和化学药剂处理后,在不同贮存温度下,观察其萌芽状况.结果表明:GA3,ABA和IAA的含量呈上下起伏的变化:萌芽前,ABA含量下降,而GA3含量上升,二者的比值随着打破休眠而缩小;在外源激素和化学药剂处理试验中,1%硫脲浸泡微型薯1 h,在25℃条件下萌芽最早,且芽长而粗;10 mg/LGA3和10 mg/L IAA+10 mg/LGA3亦能提前打破休眠;温度对促进微型薯的萌芽及对外源激素和化学药剂的吸收和转化起着重要的作用。

Abstract: Using the method of hormone ELISA,internal hormone GA3,IAA and ABA contents of potato minituber were measured at 3 storage temperatures and 4 different treatments,and the effect of the changes of GA3,IAA and ABA contents on minituber dormancy was studied.The sprout stage of minituber treated with exogenous hormones and chemicals was observed at the different storage temperature.The results show that the changes of GA3,ABA and IAA contents are undulated.Before sprouting,ABA content decreases,and GA3 content increases.And their ratio is smaller followed dormancy breakage.Among the trials of exogenous hormones and chemicals,sulfourea is the best effective.When the minituber is soaked at 25 ℃ in 1% sulfourea for an hour,the minituber sproutes the earliest and it is strongly.In the condition of 10 mg/L GA3 or 10 mg/L IAA+10 mg/L GA3,the minituber can accelerate the dormancy breakage.Temperatures play a major role in promoting the sprouting of minitubers and the absorption and transforming of exogenous and chemicals.
