Effects of Oxytropis glabra DC Poisoning on α-mannosidase of Hypothalamus-pituitary-testicular Axis in Rats

Abstract: The aim of this study was to investigate the effects of content and transcriptional expression of α-mannosidase(AMA) in hypothalamus-pituitary-testicular axis poisoning by Oxytropis glabra DC,and reveal the toxicity mechanism of O.glabra DC in future.144 male rats were randomly and equally divided into 4 groups(the control group and experimental groupⅠ,Ⅱ,Ⅲ),the dried plant of O.glabra DC was comminnuted,and different amounts of the grass powder(15%,30%and 45% the corresponding swainsonine content was 30,60 and 90 mg/kg) were mixed with the feeds in the three experimental groups.The rats consumed the forages freely until typical symptoms were observed.4 rats were selected randomly from each group every 7 day respectively,the hypothalamus,pituitary and testis were collected and the contents of AMA and its mRNA expression in different encephalic regions were measured.It was found that AMA in golgi and lysosomal expressed in hypothalamus-pituitary-gonad of all experimental groups, O.glabra DC inhibited the expression of hypothalamus-pituitary-testicular axis AMA1 and AMA2 mRNA,and the difference was obvious.The difference was not obvious in experimental groupⅠ and control group in hypothalamus-pituitary-testicular axis(P>0.05),the expression of AMA1 and AMA2 mRNA decreased in experimental group Ⅲ and AMA1 mRNA decreased in experimental groupⅡ was significant lower than control group in hypothalamus-pituitary-testicular axis(P<0.05),and it was significant lower than control group.The results showed that O.glabra DC has remarkable effects on content and transcriptional expression of AMA in male rat hypothalamus-pituitary-testicular axis,in a time-effect as a dose-effect relationship.
