摘要: 利用ISSR分子标记技术,对采自内蒙古、河北、山西、宁夏、甘肃和吉林6省区,经引种栽培筛选出的5种12个冰草属植物居群,进行DNA分子水平的遗传结构分析和评价。构建了国内冰草属ISSR-PCR扩增反应体系。研究结果表明:供试材料共检测出88条谱带,其中多态性谱带83条,多态性条带的比例为94.32%,表明供试材料遗传多态性丰富。不同物种居群间遗传多样性丰富程度不同,蒙古冰草遗传多样性最大,其次为冰草,光穗冰草最小。种间遗传分化占冰草属物种遗传多样性的56.35%,冰草物种地区间遗传多样性高于地区内遗传多样性。UPGMA聚类结果显示,同种不同材料基本能够聚在一起,但有交叉现象,部分材料表现出地域性;生态环境相似的物种遗传距离较近,聚类在一起。种间亲缘关系能够推测冰草属5个种的系统关系。

Abstract: The genetic structure of 12 Agropyron populations,including five species was analysed and evaluated by inter-simple sequence repeat (ISSR) markers. These materials were collected from six provinces in China,including Inner Mongolia,Hebei,Shanxi,Ningxia,Gansu and Jilin. ISSR-PCR amplification reaction system on Agropyron was been construct in China. The results showed that: A total of 88 fragments were amplified from 7 primers,of which 83 bands (94. 32%) were polymorphic for the test materials,and there were a large amount of genetic polymorphism in the tested materials. Populations accessions of different species had different genetic richness. A. mongolicum had the most abundant genetic diversity. The second was A. cristatum. Genetic diversity of A. cristatum. var. pectinforme was relative lower than other species of Agropyron. The genetic variation found among species was 56.35% and genetic diversity among regions is higher about A. cristatum. By UPGMA clustering,different populations of inter-species can be basically clustered together,but a few different populations of inter-species can't be clustered together. The species from similar eco-geographical situations have closer genetic distance and were clustered together.The genetic relationship among species can be speculated systematic relationship of five species on Agropyron.
