人β淀粉样蛋白基因植物表达载体的构建及转化

摘要: 人β淀粉样蛋白(Amyloidβ peptide,Aβ)是治疗阿尔茨海默氏病(Alzheimer′s disease,AD)的关键靶标之一。本试验利用重叠PCR技术获得2Aβ串联基因片段,并将其克隆到植物表达载体pBIDST(含双35S启动子和TEV增强子)中。用根癌农杆菌EHA105介导转化烟草,筛选获得了卡那霉素抗性植株。PCR和RT-PCR检测结果证实2Aβ基因已经整合进烟草基因组中并进行了转录。

Abstract: Human amyloid β peptide (Aβ)is one of the key targets in treatment of Alzheimer′s disease.The two tandem fragment of Aβ obtaining by overlap PCR technique was cloned into plant expression vector pBIDST (containing double 35S promoter and TEV enhancer).The 2Aβ gene was introduced into tobacco by Agrobacterium tumefaciens EHA105. Then the kanamycin resistant tobaccos were obtained.The results of PCR and RT -PCR indicated that the 2Aβ gene was integrated into the genome of tobacco and transcribed.
