小麦抗叶锈病基因Lr35的RGA分析

RGA ANAlysis of the Wheat Leaf Rust Resistance Gene Lr35

摘要: 根据已知植物抗病基因的NBS,LRR等保守结构域设计引物,对小麦全套抗叶锈病近等基因系材料进行RGA分析。引物对Pto-kin1IN/XLRR-INV1 从近等基因系TcLr35中扩增获得一条77 bp的特异性片段。序列分析表明,该片段与小麦基因片段Triticum urartu clone BAC 210J2,Triticum monococcum DV92的BAC克隆231A16等具有较高同源性,为Lr35的克隆奠定了基础。

Abstract: Based on the conserved domains (NBS,LRR etc) of the known resistance genes,primers were designed to amplify resistant homologous sequences in the near isogenic lines (NILs) of wheat leaf rust resistance using RGA method.A 747 bp specific fragment was found in TcLr35 by primer pair Pto-kin1IN/ XLRR-INV1.The specific fragment was isolated from the polyacrylamide gels,reamplified,cloned and sequenced. The result indicated that it has the high homology with the Triticum urartu clone BAC 210J 24, Triticum monococcum DV92 clone BAC 231A16 etc DNA fragmentas from wheat,which provide the shortcut for the cloning of wheat leaf rust resistance gene Lr35.
