摘要: 采用PCR-SSCP法对85羽绿壳蛋鸡进行促卵泡激素β亚基(FSHβ)和雌激素受体α亚基(ESRα)基因多态性检测并测序,分析其与早期产蛋性状的关系。基因FSHβ和ESRα都发现了多态,测序结果表明: FSHβ pro 2位点有3 处SNPs,分别是:A-6G,-50插入碱基A,A-05G。FSHβ exon 3有1处碱基突变,即A27G;ESRα基因5'-UTR区有处碱基变异,分别是:A-3G,T-76C,T-79C,C-102T。ESRα基因内含子1 区发现3处SNPs,分别是:T122C,A155G,C169T。与早期(36周)产蛋关系分析表明,FSHβ基因调控区的AA基因型36周平均产蛋量显著高于AB 型(p<0.05),没有发现BB型,外显子3的AA型和AB 型开产日龄呈显著相关(p<0.05),也没发现BB型; ESRα基因5'侧翼调控区的CC型和CD型36周平均产蛋量差异显著(p<0.05),而没发现DD型,其他均不显著(p>0.05)。

Abstract: Folliclestimulating hormone beta(FSHB)subunit gene and estrogen receptor alpha(ESRA)subunit gene were selected as two candidates to screen the genomic variation by means of PCR2SSCP and sequences mensuration in blue eggshell chicken(85 individuals),and the association of FSHBand ESRAgenes polymorphisms with early egg production traits were analyzed.Polymorphisms were detected in FSHB and ESRA genes,the results showed that three nucleot ide variation at A2464G,2450A insertion,A2405G,were identified in FSHBpromotor region,and a nucleot ide varia2 tion at A2447G in FSHBexon 3;Four SNPs,A234G,T276C,T279C,C2.02T,were identified in ESRA5c flanking regulation region,and three nucleot ide variation at T.22C,A.55G,C.69T in ESRA intron..Association analysis of early egg production traits showed that the egg number at 36 weeks of genotype AA was higher than that of AB(p<0.05),genotype BB was not found in FSHB promotor region;Moreover,in exon 3 site,the age at first egg was also significant difference between AA and AB(p<0.05),neither genotype BB was found.The egg number at 36 weeks was significant difference between CC and CD genotypes of ESRA5cflanking regulation region(p<0.05)in blue eggshell chicken,genotype DD was not found.Rest is no difference(p>0.05).
