摘要: 本研究借鉴经典农杆菌介导法,成功地建立了一个农杆菌介导玉米萌动胚遗传转化新体系。转基因植株经PCR,PCR-Southern,RT-PCR分子检测,结果表明外源目的基因已被插入玉米染色体基因组中,并在转录水平上表达。采用此方法可省去组织培养过程,取材方便,可操作性强。

Abstract: A new genetic transformation system by Agrobacterium tumefaciens-mediated in germinating embryo of maize was established. The result showed that integration of foreign gene into the genome of transgenic maize plants was confirmed by PCR, PCR-Southern and RT-PCR analysis. The foreign gene was expressed at transcription level. According to this method, the procedure of plant tissue culture was avoided and the germinating embryo of maize was obtained conveniently. It will provide an efficient way for genetic transformation of maize.
