猪的RBP4基因部分序列的克隆及多态性分析

Clone and Polymorphism Analysis on Part of the Pigs' RBP4 Sequences

摘要: 在猪的RBP4基因的第1外显子和第3外显子处设计引物,成功扩增了该基因的第1内含子和第2内含子,与人和鼠的同源性分别为80%和78%,说明扩增正确。采用PCR-RFLP法对大白猪、杜洛克、长白猪、民猪和杂种猪的RBP4基因进行多态性检测。结果表明:该基因经限制性内切酶MspⅠ酶切后获得3种带型:AA,BB和AB型;除大白猪与民猪、长白猪与杂种猪外,其余各猪种间RBP4基因的基因型频率分布均差异极显著(P<0.01);该基因在长白、大白和杂种猪的MspⅠ位点处于群体不平衡状态,在杜洛克和民猪处于群体平衡状态;基因多态信息含量(PIC)计算,大白、长白、杜洛克、民猪和杂种猪均表现为中度多态性(0.5>PIC>0.25),民猪的B等位基因为优势基因,推测B等位基因与猪的高产性状相关。

Abstract: Based on the exon 1 and exon 3 of the pig's RBP4 gene, a pair of primers were designed and synthesized, successfully amplified the gene's intron 1 and intron 2, and the identity of RBP4 gene sequences was 80% between pig and human, 78% between pig and mouse, the results showed that the amplification was correct. Polynerous of RBP4 gene in five strains was detected by PCR-RFLP. The strains were involved Yorkshire, Duroc, Landrace, Min pig and Crossbred pig. After genotyping with MspⅠ: there were AA, BB and AB; Except the Yorkshire and Min pig, the Landrace and Crossbred pig, the distribution of other pigs' genotype frequencies were significance (P<0.01). This MspⅠ site in the gene in Landrace, Yorkshire and Crossbred pig population were disequilibrium, in Duroc and Min pig populations were equilibrium. Used the polymerous information content (PIC) to calculate, the five pig breeds appeared normal polymerous (0.5>PIC>0.25). The allele B of the Min pig is advantageous. It is presumed that the allele B is associate with pig's high production character.
