摘要： 将“津引薯8号”马铃薯薄片和含有CaMV35S启动子控制的类甜蛋白（TLP）基因与紧密连锁的Ubiquitin启动子控制的bar标记基因的农杆菌双元表达载体进行共培养。切下马铃薯薄片表面产生的幼芽，用含有除草剂bialaphos的MS培养基进行初步筛选。当获得的生根抗性芽生长成株后用TLP蛋白抗体和TLP基因特异引物对成株进行间接ELISA检测和PCR检测。两种检测结果一致率为90%，推断以bar基因为筛选标记、紧密连锁的外源类甜蛋白基因已经进入马铃薯基因组并得以表达。

Abstract: Tuber discs of "Jinyin potato No 8" were co-cultivated with an agrobacterium tumefaciens binary vector, carrying both the thaumatin-like proteins gene under the control of the CaMV 35S promoter and the bar gene as a selectable marker under control of the ubiquitin promoter. Tuber discs started to form a number of shoots on shoot inducing medium two to three weeks after co-cultivation. The shoots were inoculated on MS medium containing the herbicide bialaphos. The potato seedlings which grew well on MS medium containing bialaphos were assayed by indirect ELISA. The positive samples were assayed by PCR to see the existence of the TLP gene. The results of it indicated that the foreign thaumatin-like protein gene was introduced into the potato genome and expressed in the transgenic potato.
