摘要: 本文应用垂直板不连续系统的聚丙烯酰胺凝胶电泳(PAGE)法,分析了5个杂交一代种(佳粉一号、佳粉二号、佳粉十号、佳粉十五号、双抗二号)发芽种苗的五种同工酶(苹果酸脱氢酶(MDH)、乙醇脱氢酶(ADH)、磷酸葡萄糖变位酶(PGM)、酯酶(EST)、异柠檬酸脱氢酶(IDH)).发现5个杂交一代种中,3个杂交一代种与其双亲的苹果酸脱氢酶(MDH)谱带有明显稳定的区别。两个杂交一代种与其双亲的酯酶(EST)谱带有区别。在此基础上,建立了用苹果酸脱氢酶电泳分析检测番茄一代杂交种纯度的技术,检测结果与田间鉴定结果基本一致。

Abstract: F iv e isozyme system s, namely dehydrog enase ( M DH ) , alco ho l dehy drog enase( A D H ) , pho spho gluco mutase ( P G M ) , est erase iso zym e ( EST ) and isocit ric dehy drog enase( IDH ) in ex tr act s f rom cot yl edons of germ inat ed t omat o seeds w ere analysed by polyacry-lamide gel elect ro phoresis ( PA G E) . Poly morphisms w ere observed at t hree loci ( MD H-1,M DH-2, MD H-3) . T he purit y of F1 hybrids , Jiaf en 1, Jiaf en 2 and Jiafen 15, w as t est ed by utilizing M DH-1 loci as markers, and the r esult s co mpar ed fav orabl y w it h t hose obt ained fr omfield purit y ident ification.
