A HPLC Method for the Determination of SQ in Plasma and Tissues of Animal: Regression Analysis within the Drug Contents of Various Samples

摘要: 采用高效液相色谱(HPLC)分析法测定动物血浆和组织中的磺胺喹恶啉,其准确度与专属性好,最低检出限为1×10-7g.色谱条件:以十八硅烷磺酸钠(ZorbaxODS反相柱)为固定相,紫外检测波长254nm,流动相为甲醇:水(50:50),流速为1ml/分,磺胺二甲基嘧啶作内标物.通过血浆与组织SQ含量间的回归分析,还建立多种回归方程,为组织中药物含量的间接预测提供计算依据.

Abstract: A HPLC method for the seperation and determination of sulfaquinoxa-line(SQ) in plasma and tissues of animal is reported. This method exhibited very high sensitivity and selectivity. The minimum detectable quantity is1×10-7g with high accuracy. The chromatography fitted with reversed-phasc Zorbax-ODS(Octadecylsilam)column and equipped with a 254 nm UV detecter. The mobile phase was a mixture of methonal. distill water(50/50,v/v) with a flow rate of 1 ml/min. Sulfamethezin was used as a internal standard. According to the regression analysis between drug levels in plasma and SQ contents in tissues, the various regression equations were established and the calculation basis were provided for the indi-rective prediction of drug levels in the tissues.
