Several preparation methods for Exserolalum tureicum intact chromosome DNA were comparatively stuied, in order to establish an all-sided system to analysis the molecular karyotype of this fungi. The results showed that chromosome DNA of Exserohalum tungzaghubum were only obtained by the agarose-embedding protoplasts method, other methods gave incompetent results. Moreover, two chromosomal bands were separated by PFGE, both were more than 2200 kb.
