Cytological Observation and Biochemical Analysis of Maize Root Meristem Subjected to the VHZ Stress

Abstract: Studied the VHZ′s toxic effect to maize meristem in cell and biochemistry level. VHZ is a microtubules depolymerization triazole compound. Experiment results revealed that prophase,metaphase synchronization division cells, chromosome structure variation (chromosome bridge,chromosome sluggish,chromosome condensation) could be induced by VHZ in root meristem of maize. The threshold concentration and time were 20μmol/ L for 12 h. Synchronization division cells and chromosome structure variation were thought to be directly related to the disturbance of spindle function by the immunofluorescence test. Two protein species were disappeared and the content of four protein species were reduced in root meristem by 2D SDS2PAGE analysis. They could play an essential role in cell defence mechanisms against VHZ.
