Application of Microsatellite Markers in Endangered Hucho taimen Enhancement and Releasing

Abstract: In order to study the application of microsatellites markers in endangered Hucho taimen enhancement and releasing hucho releasing,eighteen microsatellites markers were used to analyze the genetic diversity and structure of release-recapture population (JC),Tahe River population (TH) and released parents' population (HT,YZ) in the present study.The recaptured individuals were also identified.The results indicated that Na,Ne,I,He and H indexes exhibited the same tendency,HT > JC > TH > YZ.The observed heterozygosity (Ho) was highest in YZ population while lowest in TH population.All of these four populations possessed a medium level of genetic diversity.AMOVA analysis showed that there was no significant difference within four groups while extremely significant difference between populations.The genetic similarity was highest between YZ and HT population with the index of 0.718 3,medium-level between JC population and other three populations with the index of 0.566 5-0.689 5, and lowest between YZ and TH population with the index of 0.495 9.The results showed that we could understand the genetic background of the parents and assure the released descendant's polymorphism by microsatellite markers.The released individuals were identified by microsatellite markers.In conclusion,this study provided the basis for the next stage of artificial releasing work.
