Abstract: The results showed that N6 basal medium supplemented with 5% sucrose,0.8% agar,0.5 mg/L NAA and 1.0 mg/L 2,4-D facilitate plantlet induction of unfertilized ovule culture in summer squash(Cucurbita pepo L.).Diploid plantlets, which account for 60% of the total plantlets by taking account of chloroplast number in guard cell were confirmed of pure lines through investigation of agronomy traits of selfing offsprings. That means the diploids were pure line and they originated from gynogenesis of haploid megaspore cells.

Key words: Summer squash(Cucurbita pepo L.), Unfertilized ovule, Double一haploid
