Abstract:
A novel and practical dot blot hybridization assay was developed for detectionand identification of inonocytogenes.The biotinlabeled synthetic oligonucleotide probe wasconsidered specific for the listeriolysin gene of monocytogene.Using the probe,we wereable to detect as little as long of DNA and as few as 150 bacterial cells in the simulatedsamples cultured overnight.It reacted with all 12 strains of L.monocytogene.stested,and nocrossreaction with other species of steria and bacteria took place.Dot blot hybridizationgot more positive results than bacterial cultures in detection of Lmonoc ytogenes in the porkand milk samples obtained from the market.It is concluded that this method is applicable todetect monocytogenes in animal food.
