In Vitro Culture of Immature Tassels of Zea diploperemis and Its Interspecific Hybrid.Ⅰ.Direct Formation of Adventitious Buds and Plant Regeneration

Abstract: Plants were regenerated with high frequency from immature tassels of Zea mays L.(maize inbred 330),Zea diplopetennis,and Zea maysZea×Zea diplopennis hybrid cutured in vitro.Adventitious buds and shoots were directly developed in the primary culture from the immature tassels cultured on the medium MS containing 2 mg/1 6-BA.Subcultured on the same medium,rapid,clonal bud and shoot propagation was obtained.The buds and shoots were very easy of rooting by transferring them to root-inducing medium,MS containing 0.5 mg/1 NAA,300 mg/1 lactalbumin hydrolyzate(LH),and 1000 mg/1 activated charcoal.The developmental stage of immature tassels and the combination of phytohormones were most important for successful culture establishment.The application of this technique was discussed.
