一株牛源醋酸钙不动杆菌的分离鉴定、药敏试验及耐药基因检测

Isolation, Identification, Drug Sensitivity Test and Detection of Drug Resistance Genes of Bovine Acinetobacter calcoaceticus

摘要: 为研究醋酸钙不动杆菌的致病机制及防控措施,对疑似患有醋酸钙不动杆菌的病死牛肺脏组织进行病原分离,得到了1株优势菌命名为9-1。对分离菌株进行形态学观察、生化试验、16S rDNA基因序列测定,动物致病性试验和药敏试验。结果显示,分离菌株为革兰阴性杆菌且对试验小鼠具有较强的致病性,经生化试验和16S rDNA基因序列测定分析确定为醋酸钙不动杆菌。药敏试验结果显示该分离菌株对氟苯尼考、头孢噻肟、头孢氨苄、阿奇霉素耐药,对其余8种测试药物敏感。经β-内酰胺类(TEM、DHA、OXA-23、OXA-58)耐药基因检测结果显示均为阴性。为醋酸钙不动杆菌引起的疾病选择合理的抗菌药物提供了一定科学依据,并为进一步研究其耐药机制奠定了基础。

Abstract: In order to study the pathogenic mechanism and prevention and control measures of Acinetobacter calcoaceticus, the pathogen isolation of the diseased dead bovine lung tissue suspected of having Acinetobacter calcoaceticus was obtained, and one dominant bacteria was named 9-1. The isolates were subjected to morphological observation, biochemical test, 16S rDNA gene sequence determination, animal pathogenicity test and drug susceptibility test. The results showed that the isolated strain was Gram-negative bacillus and had strong pathogenicity to the test mice. It was identified as Acinetobacter calcoaceticus by biochemical test and 16S rDNA gene sequence analysis. The drug susceptibility test showed that the isolated strain was resistant to florfenicol, cefotaxime, cephalexin, and azithromycin, and was sensitive to the other eight test drugs. The results of β-lactam (TEM, DHA, OXA-23, OXA-58) drug resistance genes were all negative. This test provides a scientific basis for the selection of reasonable antibacterial drugs for diseases caused by Acinetobacter calcoaceticus, and lays a foundation for further research on its resistance mechanism.
