大豆gma-miR160o启动子的克隆及植物表达载体构建

Isolation of gma-miR160o Promoter from Glycine max and Construction of Its Plant Expression Vectors

摘要: 为了研究miRNA表达的调控机制,以大豆Williams 82基因组DNA为材料,根据预测的 gma-miR160o 启动子序列设计引物,采用PCR方法克隆 gma-miR160o 上游一段长度为1 910 bp的启动子序列。采用PlantCARE和PLACE启动子在线预测工具分析表明, gma-miR160o 启动子序列具有TATA-box、CAAT-box基本的顺式作用元件和一些参与非生物胁迫、光和植物激素应答相关的顺式作用元件。将 gma-miR160o 启动子替代pBI121载体中的CaMV35S启动子,构建了与 GUS 融合的启动子表达载体。

Abstract: In order to investigate the regulation mechanism of miRNA expression, in this study, based on the predicted sequence of gma-miR160o promoter region, primers of promoter sequences were designed.A 1 910 bp fragment was cloned from the genomic DNA of the soybean Williams 82 by PCR method.Promoter sequence analyzed by the database of PlantCARE and PLACE showed that the sequence contained basic elements TATA-box, CAAT-box and some cis -acting elements involved in response to abiotic stresses, light and plant hormones.The genomic DNA fragment in gma-miR160o promoter and GUS fusion expression vector were constructed by replacing CaMV35S promoter of pBI121.
