黄瓜基因组DNA提取与RAPD分析

摘要: 采用SDS法、尿素法、改良SDS法、氯化苄法和CTAB法对黄瓜的基因组DNA进行提取.结果表明,改良SDS法提取的黄瓜叶片DNA具有典型的天然DNA分子的标准紫外吸收光谱特点,其A260/A280在1.7~1.9之间,DNA的产量为50 μg/g.用该法提取的黄瓜DNA适合进行RAPD分析.

Abstract: The preparat ion of DNA samples with good quality is the basis for researches of plant molecular biology.The DNA samples of cucumber male sterile line YS and maintainer line MC were prepared by SDS, Urea, Improved SDS, Benzyl chloride and CTAB method, respectively. The results showed that DNA samples obtained by improved SDS method possessed the standard ultraviolet absorbance spectrum of the pure natural DNA and the A260/ A280 was between 1.7- 1.9, the yield of DNA obtained by improved SDS method was 450Lg/g young leaves, and the DNA samples prepared by this procedure were suitable for RAPD analysis.
