尖裸鲤5种组织乳酸脱氢酶和苹果酸脱氢酶的电泳分析

Electrophoretic Analysis on Lactate Dehydrogenase(LDH) and Malate Dehydrogenase(MDH) in Five Different Tissues of Oxygymnocypris stewartii

摘要: 为了了解尖裸鲤的生化遗传特性,丰富尖裸鲤种质资源研究的内容,通过配制不连续浓度聚丙烯酰胺凝胶,采用聚丙烯胺凝胶垂直板电泳对尖裸鲤5种组织(心脏、眼睛晶状体、肌肉、肝脏和肾脏)中的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)进行了电泳。结果表明:所测样本鱼心脏、眼睛晶状体、肌肉、肝脏和肾脏中的LDH酶带数分别为:12~16条,17~18条,6~13条,6~10条和12~17条;MDH酶带数分别为3条,4条,3~5条,2条和4~7条。LDH在尖裸鲤厌氧器官(骨骼肌和肝脏)中表达的酶带数总体少于非厌氧器官(心脏和肾脏)。尖裸鲤的MDH分为上清液型(s-MDH)和线粒体型(m-MDH)。尖裸鲤的LDH和MDH同工酶系统具有组织特异性,尖裸鲤相对较多的LDH酶带数可能与其独特的栖息环境和A亚基与B亚基的变异共同作用有关。本研究可从生化遗传角度为尖裸鲤的资源保护和利用提供一定理论依据。

Abstract: The purpose was to learn about the biochemical heredity characteristics and to enrich the research data about the germplasm resources of Oxygymnocypris stewartii. By preparing concentration gradient polyacrylamide and using the vertical polyacrylamide gel electrophoresis,lactate dehydrogenase(LDH) and malate dehydrogenase(MDH) from five tissues(heart,crystalline lens,muscle,liver and kidney) of Oxygymnocypris stewartii were preliminarily studied.The results showed that:the band numbers of LDH isozymes from heart,Crystalline lens,muscle,liver and kidney were 12-16,17-18,6-13,6-10 and 12-17 respectively,and MDH isozymes 3,4,3-5, 2 and 4-7.The band numbers of LDH isozymes expressed in anaerobic organs such as muscle and liver were less than that in non-anaerobic organs such as heart and kidney.MDH isozymes in Oxygymnocypris stewartii were divided into supernate type and mitochondria type.LDH isozymes and MDH showed tissue specificity.More bands of LDH isozymes in Oxygymnocypris stewartii might be related to the interaction between it's unique habitant and the variation of subunit A and subunit B.The results could provide theoretical basis for the conservation and utilization of Oxygymnocypris stewartii from the biochemical genetics level.
