花生黑斑病菌致病毒素提取及其活性测定

Activity Analysis and Extraction of Cercospora personata Toxin

摘要: 为了明确花生黑斑病菌的致病因子及致病机制,对该病菌分泌的毒素进行了提取和生物学特性测定.通过对花生黑斑病菌毒素产生条件筛选和优化,采用氯仿提取、硫酸铵和柱层析分离纯化病菌产生毒素.结果表明:花生黑斑病菌在V8培养液、光/暗交替、静止条件下培养20 d产毒活性最强,用等量的氯仿提取,再经过80%饱和度的硫酸铵沉淀透析和柱层析纯化,可以获得较高活性的花生黑斑病菌毒素.初步分析显示,毒素由A和B 2种成分构成,但仅A成分对花生有致病性,B成分没有,A成分是毒素的主要成分;另外,病菌毒素具有一定的热稳定性和较强的极性.

Abstract: For the purpose of expliciting pathogenic factors and pathogenic mechanism of Cercospora personata,this paper extracts toxin secreted by Cercospora personata and analyzes the biological characteristics.This paper filtered and optimized the produce conditions of toxin.Then the germ toxin was purified by chloroform extraction,ammonium sulfate purification and column chromatography isolation.The results showed that the activity of Cercospora personata in V8 nutrient solution which exposed to light and darkness alternatively and cultivate under static conditions for 20 days was the highest.Higher activity Cercospora personata toxin were obtained by extraction with equal amount of chloroform,sedimentation dialyses with 80% saturation ammonium sulfate and column chromatography purification.Preliminary analysis showed that the toxin was composed with two components.Component A was pathogenic to peanuts while components B was not.Component A was the major components of the toxin.Furthermore the germ toxin had a certain degree of thermostability and strong polarity.
